I would pick the G35.
1.) Manual Transmission
2.) Larger Interior Space
3.) Real aluminum trim vs. painted plastic.
4.) Better looking IMO.
5.) Newer than the IS.
6.) More of a drivers car.
7.) 4-Wheel Steering.
8.) More mods available.
9.) Better warranty.
10.) Price.

id pick the G35 easily for the same reasons as DrUnBiased.
mainly the G comes in stick, has a bigger interior (the IS feels cramped to me), is better looking, is more of a drivers car, and has price advantages

I was in this dilemma two weeks ago, and ended up with a IS350. I think they are very close competiters and very very hard to decide which one to get. I just like IS's styling a bit more that is why I bought one.

1) A more refined and practical automatic.
2) I did not need the extra room in the back (solo driver 95% of the time).
3) My wood trim is real wood.
4) Better looking IMO.
5) G35 engine is older. IS350 engine is newer.
6) More of a luxury driver's car. (think of the new GS commercial; just because I can, doesn't mean I should...)
7) RWD is fine for me. Gas mileage is better.
8) I'm quite happy with all the mods I've done to my car. Mods are out there...
9) Warranty is both 48 months, with 10k miles difference. If you lease/drive less than 12k miles a year, there is no difference between the two.
10) Resale is better.
Now, other people may not choose the IS because of my reasons, but I did find the IS a better fit for me than the G35...

I'll choose IS350, simply, because it's big L.
As long as Lexus can keep introducing high end cars like LS600HL, and keep its whole products quality not drop, its prestige will grow up, and its low end products should win automatically.

I don't have a deep affliction for either, I would buy a german aside of those two choices.... however given this specific choice I'd take the "L" for it's overall quality and driving experience. I have never gotten the bad taste out of my mouth from Nissans years of skimping on cheap plastic interiors and the wear and tear always shows on a nissan first and a Lexus last.

I haven't driven either so I can't talk about dynamics but...
I'd pick the IS because:
looks better (completely dependent on buyer)
RWD FTW!
auto with paddle shifters are good enough in this category
relaxed interior-the G's interior (drivers seat) makes me claustrophobic
ELECTROLUMINESCENT GUAGES!!!!
ERGONOMICS ERGONOMICS ERGONOMICS. Everything is where I think it should be in the IS where as the G..."where are the $%$#'n sunroof controls?"

Looks are subjective so I wont state which I prefer.
G35 also is RWD, or AWD option.
G35 also has paddle shifters with the sports package.
As for claustrophobic interior, I'm only 5'4" and my wife is 5'2" and the IS felt very small and tight for us. Even with my small stature, adjusting the drivers seat to my liking then sitting in the back seat was tighter than I would prefer. I really don't know how taller people can sit in the back of the IS. The G had much more room. I think the IS has the claustrophib interior.
The one thing I think is a major plus for the G35 is the LCD screen comes standard with or without navigation. All G35s also come standard with an Audio and Video AUX input.
