shift gear in auto transmission vehicle
Are we supposed to lift the foot off the gas pedal while changing gear up and down in an automatic transmission vehicle in manual mode?

In most cases, when up-shfting manually, it isn't necessary, unless you are lead-footing it. In some cars, the transmission's computer eases up on the engine's torque automatically as you up-shift, no matter how hard your foot is on the gas, to prevent excessive shift-shock to the transmission. Downshifting manually, though, if the transmission doesn't hame automatic rev-matching on the downshifting (most transmissions don't), I'd give it a small amount of extra gas (similar to heel-and-toeing on a traditional clutch and manual-transmssion) before moving the shift lever or paddle, to make sure the engine speeds up a little during the downshift and is close to the transmission's speed.....that will lessen the abrupt increase in engine RPM that comes with the downshift upon engagement.
