I need suggestion regarding 4 speed and 5 speed transmission .

_ My understanding is that 1997 es300 comes with 4 speed transmission

_ I never counted the gears while I drive , until today

_ When I release the break the trany is in gear 1

_ After that I counted it goes all the way to 5

_ I can only feel shift from 1 to 2

_ After that the shift is not noticible unless you see the trachometer

So now my question is

1- Do some 1997 es 300 comes in 5 speed ?
2- Or the previous owner changed the trany to 5 speed ?

No, you can't count.
You're probably counting when the torque convertor locks up if you use the tach.

Except for the rare 5-speed manual transmission in some of the '92-'93 ES, all of the ES are either 4-speed auto w/ overdrive ('92-'01 ES) and 5-speed auto ('02-'06 ES).
