1995 ES300 Tranny issue

New to the forum. I need some help. My 1995 ES 300 with 284K (kids car) is acting weird. I was having transmission problems, (I thought) and replaced it with a used one. The same symptioms were there after the switch. The car seems to be stuck in drive when I take off, and would occasionally cut off. Kinda like if you were starting a manual transmission in 3rd gear. If I start out on low gear and manually move through the gears no problem. The CEL is on with both a P500 and P720 code. I changed out both the sensors myself on the transmission and the problem is still there. The weird thing is that occasionally the RPM's spike (like the car is going out of gear) when the lights are on. I used the daytime running lights as a general rule, but no longer. I have reset the computer by unhooking the battery cables and the problem persists. I am not real car savvy, but can do some minor things. I believe I have some type of electrical problem, and do not know what to do. Also, occasionally I get a blinking OD light, (not rapid) and I have no overdrive at present. The OD did work one time recently, but that's it.
I have no problem operating the car myself, and haver very little money in it, but obviously do not want to hand it back to the kids without it being more relaible. I did have the tranny flushed after the used one was put in, and it does seem to shift better but the major issues remain.
Does anyone have any thoughts or suggestions? Thanks.

Both of these codes relate to the vehicle speed sensors. This WILL effect your transmission shifting, and could cause the problem you described.
The problem could be the sensor itself, or just as likely it's associated wiring.
