Been reading that some are experiencing excessive wind noise at highway speed. Is this noise due to the lines of the body's construction around the windows? Or is it due the window(s) not being flush against the weatherseals of the door?

Welcome e-Mel. We know that if the door is the rubber seal is "shimmed" around the window this can reduce the sound issue coming from the front portion near the 'A' Pillar. We also know that if you tape up the outside of the window to the door itself then that also pretty much kills the noise. So the current thoughts on this are that its a door rubber (or weatherseal) issue.
Some people don't have this problem. This tends to suggest it is a manufacturing issue and not a design defect. There is no official Lexus fix at this time. Lots of threads though on this here at CL.
