You need to pop that cover off/out.

Right behind that is the clear color plastic housing tray which contains the actual cabin filter.
You have to release the latchs and the tray will then slide out.

Voila !!! There is the filter.
When placing the filter in, just keep in mind the filter will have "orientation arrows" showing which side faces up.

After that, just reverse the process.

In case you're interested, I got the filter at the Toyota dealer.
PART number is 87139-07010
Had to be cheaper than the lexus dealer.

Good luck and Happy Motoring !
Thanks for the information. I went to a Toyota dealer (Toronto, Canada) to get the cabin air filter for $35 and it only takes a few minutes to change the old one.

Mine was changed for the first time since new (9/06 - 22,000 miles). Was filthy / dirtier than the engine air filter that I changed also.

I just replaced mine with a TYC filter purchased on Ebay. I want to caution others that on the original filter there are arrows with the word "up". On the TYC filter there are arrows with the words "air flow". If you read the instructions with the TYC filter they tell you the air flow is down. Therefore the TYC filter should be installed with the arrows down.

This is obvious if you read the instructions, but many people do not.

A word of caution; I just replaced my '07 ES350 cabin filter with a '07 V6 Camry filter, got the filter home and to my surprise they are different! The Camry filter has fewer pleats, the pleats are farther apart and it was made of a different material. Sure enough, I called my local Lexus dealer and the part numbers are different. I took the Camry filter back to Toyota where the parts guy did a little checking, and found that the '07 and later Avalon filter is in fact the same as the ES350 cabin filter. Cost me 32 dollars instead of 44 at Lexus and now I feel better. The Camry filter fits and would probably work fine, but I wanted the correct filter.

I will now be checking the cabin filter on my 2007 ES 350,but was actually looking for information on how to replace the engine air filter,as the manual is not helpful.Has anyone tried it?I was quoted $30 by an independent garage who said I needed one.Maybe this is reasonable but I have easily done it myself on other vehicles. Thanks for any info.

The ES350 is probably the most difficult air filter I have dealt with but if you've been doing it for years there is no reason you can't handle this as well as a shop. It' the same procedure as always, release a few clips and replace it - it's just a tight fit on this car.
