I recently bought an '07 es350 with low miles. I've noticed that at times I can hear a very faint sound that is like a mouse running on wheel from my front driver side... almost seems like it's coming from the wheel area. Weird thing is that when I put the window down I can't hear it, only when the window is up. It happens more often when go over bumps. It's really been bugging the heck outta me and I'd love to get this figured out.
ANY help is HIGHLY appreciated!

check tires for any rocks, nails or screws that might have gotten stuck in it. that's my advice.
anything beyond that is a mechanical problem. IE. wheel bearing, suspension, maybe something rubbing against the brakes or brake shield.. etc. ... if you still have a warranty on the car, take it in to get it checked out.
if not, i certainly hope you have some car savy friends at your disposal who can take a quick peek at your vehicle.
