Does anyone know when the next generation of the ES350 is going to be available in dealerships?

Will this have the hard drive based navigation with the newer style interface and hard drive to load MP3s?

Strictly an uneducated guess. The 2010 model was cosmetically modified, along with the Nav and interface. I suspect the next generation with be for model year 2013, which will be released in 2012.

Does the 2010 ES have the option loading MP3s and sorting them for playback?

No, the 2010 does not have a hard drive for music storage, just a USB port for Mp3 players or memory sticks.
