On my 2008 ES 350, the smart key works almost completely fine. Doors unlock when the handles are grabbed, and the vehicle starts fine + proximity sensors.
However, on the driver's door only, the button to lock the car does not function. The door will lock and unlock with the key fob fine, and the door will unlock with the smart key access by grabbing the handle. Only this button does not function on the driver's door. The other buttons on the other three doors work fine and lock the car properly. Could this be a bad fuse or will the door handle need replacing?

I doubt a fuse. I'm not aware of a fuse that would only affect one single button. My guess would be that the button on the driver's side door handle is bad. The button is just a switch and switches go bad. Especially exposed out in the open.
