Just an update for completeness (so those in the future might find this helpful), the gush of wind noise is predominantly attributed to missing floor pan underbody plugs (4 total in my case), see the following post for complete info:

Among other things, pulling up the rear drain tubes in the trunk and securing with rubber bands tied around them (as mentioned in this post) will also help to minimize the wind noise--esp. if your drain tubes are sticking more than a few inches down into the rear wheel well. You should leave exposed about 0.75" of the drain tube to the wheel well.

Further, replacing my Bridgestone LM-22 winter tires for Michelin Pilot Sport A/S helped with reducing the road noise.

However, the most appreciable and important fix was replacing the missing rubber plugs in the floorpan underbody holes. The unplugged holes were perfect places for allowing road noise and wind noise to penetrate into the car's cabin (esp. the passenger front hole due to it's forward angle).

After the above fixes, the car is significantly quieter (less road noise and also the gush of wind sound is almost nil)--thanks to all for your help.

I realize this is an old post, but rather than starting a new one and going through this again, it may be best to continue this one.

I too have had a bit of wind noise in my 2001 GS430 that seems a bit excessive. It sounds like common culprits are:

1.) Front and rear floor-board rubber plugs missing

2.) There is technical service bulletin from Lexus about the rear view mirror sealing causing noise

3.) Sun-roof drains (in trunk) sticking too far below the surface of the body? Still not sure what the theory is behind this?

4.) Weatherstripping around doors/windshield

I will check the plugs first and see how they look.
