My wife and I took the 450H on a 300 mile cross state trip this weekend, and with the cruise set at 70 and the AC running full blast, mileage clocked in at 31.5mpg. I manually calculated the milage to double check the trip computer, and it was spot on. The EPA highway rating is 28mpg, so I'm quite pleased.
As for city mileage, I only averaged 21 over the first 400 miles of suburban driving, which is well below the EPA estimate of 24. I was getting 20mpg in the GS 300 driving the same routes, so this is disappointing.

GS 450H = 21 city / 31 hwy
The part that's dissapointing is the city mileage. Driving the 450H the same way as I was drivng the 300 - not faster, not slower, not more agressively - then I assumed the hybrid would get better mileage than the 300 in city driving. My daily driving is fairly conservative under conditions with frequent stops and speed limits in the 30-50 zone. This should be where the hybrid technology pays off, but so far I haven't seen that.
I know mileage varies greatly from driver to driver, and this is why I compared the 450 to the 300. The fact that the 450H can do 0-60 2 sec quicker is a real plus, but I think if you put that extra potential into frequent use, you're going to get milage well below what I've seen so far. I'll put the car in sports mode on the next fillup and push a little harder to test my theory.

I bought my GS450h in mid January of this year, and was initially getting about 22mpg on my regular commute (which is about 15 miles long, with about 9 of it being a semi-congested highway). But then spring and summer came, and my mpg increased dramatically to over 25 mpg.
On the highway, having cruise control set for 69 mph, I get about 31-32 mpg, depending on whether there are any hills or not.
