Keyless start and locking problem!!!

I had the same problem with my 06 GS430 on the second day I had the car. I parked at a restaurant and couldn't get the car to lock with the rubber button on the doorhandle. I got back in the car and turned the lights off in case they were going to stay on. After I closed the door, the rubber button worked and the doors locked.
When I came out after dinner, the car would not unlock by touching the door handle, and the lights under the mirrors were not illuminating. I sat on a bench next to the car trying to figure out what to do (and feeling like an idiot!!), then finally called a friend who is a manager at my Lexus dealership, and he informed me about the metal key (which I hadn't known about) and I had to hold the key fob up to the start button just like you did because it wouldn't start normally. I took the car to the dealership the next day, but there were no problems found, and no mention of a TSB at that time.
It hasn't done that again, but one time at the same restaurant, it was slow to lock or unlock just for an instant, so I feel there must be some kind of interference in that shopping center. I live right in the heart of Dallas...not near any military bases that I know about.

Happened to me several times. I live in San Diego and every time I go to the vista point of Mt. Soledad (the cross) in La Jolla, I know I will not be able to turn the car on unless I put the FOB close to the ignition button. Just too many microwave towers around. Also, I had the problem recently due to the battery in the FOB running out, after about a year of operation.
