18inch wheels / 245-40 summer tires

How is the ride with this option?

Does it make the ride much harsher, or is there little noticable difference between it and the stock 17 inch wheels and tires?

You don't have any problem. That is the configuration that comes stock on GSs equipt with 18s from the factory.

I test drove the GS450h, which comes with 18" wheels. Very smooth, I did not notice any harshness in the ride.

In comparison it is a little harsher.

I actually like the ride of my 18" summer set up better than my 17" runflat set up. Now, this fall when the runfarts get replaced, it may be another story.
