Do this.
In the middle of the night, when nobody is on the road, drive to the nearest highway onramp. Fill up at a gas station as near to the onramp as possible so you have a full tank and are starting from scratch.
Get on the onramp, accelerate to the posted speed limit quickly, then engage the cruise control. Drive for half hour or more, then stop and look at what the computer is telling you.

No disrespect to the OP for I do not know you, but it has to be your driving style effecting your MPG. Your view of 'conservative' driving may be someone elses 'bat out of he11' When I travel to the Wash DC area to visit family, I have to change driving style by going much faster than I normally would just so that I do not get run over. My MPG's drop because I am having to drive much faster. Now in my 07 Tahoe, no matter how I drive it, I will always get 14-15mpg in town, 19-20 on the hwy.

I have a 2009 GS 350 and get only 13-14 mpg in city driving, and lucky to get 17 mpg on highway driving. I use premium gas and have tried various brands.
My car was checked several times by Lexus and they said that they found nothing wrong.
My 8 cylinder GS 430 got over 18 mpg in city and 27 mpg on the highway. How come this new SIX cylinder car does worse than a EIGHT cylinder one?
Any thoughts?

Have you measured the fuel efficiency manually? If theI wonder if the computer is wrong.

Change your oil.
What percentage of city driving do you make on a daily basis? This will determine if you need to change your oil more frequently. Also, I don't know what kind weather you guys have in Florida during the winter, but the engine will require more fuel to reach optimum operating temps in areas that are colder.

Here's a couple of things I would try:
1. Disconnect the battery for about 1/2 hour and then reconnect it. I believe this will clear the currently "learned" fuel injection settings, and force the car to relearn them.
2. Make sure the car is truly getting into lower gears while the tranny is in "D", which I presume it is. If the car was staying, even in 4th gear, that could affect the MPG to a good extent.
If none of that worked, I'd get corporate Lexus involved and bug the crap out of them until they got it fixed.

I have the exact same issue with the 07 GS 350...tried all brands of gas, ECU reset, etc, but nothing worked. Very conservative driving/acceleration.
My other car, which is rated to use more gas, gets 1 MPG better than the rated fuel consumption...and I actually press the gas/rev more aggressively since its slow to start.
Dealer won't do anything, saying its "normal."

you may want to consider looking at BOTH your calculations combined with the car's computer. They are likely to be very close but then you could at least learn to trust the computer or not, based on the results.
My 07 GS350awd gets approx 16-20 around town. I have been pressing the gas heavily lately, especially in (safe) corners to test the car in the snow.
Like you, I was surprised to see the mileage to be not improved over my GS400.

I have a 2010, and right from the start I have gotten great mileage. In a mix of about 85% highway (in the Houston area) and 15% city I easily am getting around 26mpg. Even the city driving does not pull down my mileage as much as past vehicles have.
I hate to ask stupid questions, but you are not driving with the gear selector in "sport" mode, are you?

Just to add onto this train, I had a 2006 gs 300 and got roughly 25/30 and now have 2008 gs 350 and get 18/25 in los angeles traffic. On the wide open road, say interstate 5 to San Francisco or 15 to Vegas I can still get 30 mpg in the 350. And thats averaging 90+ mph. Good luck

Have you calculated the numbers? Assuming that you are using whole tank for 300miles which is 18.8 gallon that gives ~16miles per gallon or better if you leave some before refueling... so it is not 13... meaning something wrong with your computer/sensors.

14 to 16 mpg was typical in my stop light to stop light in town driving on my 07 GS 350.
You can switch to view your "current" fuel consumption results by using the "DISPLAY" switch on the steering wheel. That will really give you an idea about where or why your use of fuel is high.

but idling kills mileage, it's hard to get a good average when you're getting zero mpg. that's why i am surprised the GS 450 hybrid does not do better on the city cycle..
