My 06 GS430 does automatically lock when I put the car in drive, and unlocks when I put the car in park. It's actually kind of annoying, and I'm concerned that the door lock actuators could wear out prematurely because of this. I'm taking the car in tomorrow for an oil change and am going to ask them to disable this feature.

Check your manual, it should have two sets of instructions, for power button on/off and shifter position.
I did teach my GS 430 to close doors once in drive and open when in park. That’s as much as I remember since 2008, hope it helps.

mine will look when drive, but it won't unlock when put in park. I can pull on the latch inside and it will unlock and open the door all in one. This only work for the two front doors, the back doors they have to unlock and pull the latch to open the door.

Thanks to this thread, I was able to disable the locking/unlocking feature on my own. It's a little tricky, but if you follow the steps in the manual, it can be done. I don't know if this made a difference, but I tried doing it a few times without the parking brake on and it didn't work. Once I applied the parking brake, I was able to make the change.
