2006 gs300 manifold removal
Anybody ever removed them or replaced them before?
Any instructions on how to removal them?
Planning to buy some headers and need instructions on removing the stock manifolds...wonder if its hard?

You should be able to find removal/installation instructions at techinfo.toyota.com. As for whether it's hard or not, that really depends on how comfortable you are with wrenching on cars and how extensive your tool collection is. There probably won't be a huge amount of room to maneuver, but it's not an inherently complicated job. Be sure you have an O2 sensor socket.
On the other hand Othic is a Lexus tech, so I'd take his advice into consideration unless you're confident you can do the job

Don't take what I said the wrong way, I'm definitely not trying to discourage you from doing it. The first piece of auto repair I ever did (aside from oil changes with my dad when I was 8) was the water pump on my 3GS – it wasn't an easy job ad it took like 6 hours, but it turned out fine! Replacing the water pump is probably a more technically challenging DIY than headers are. The point is, if you have a good set of tools, good instructions, some free time, and are patient, you'll be able to do it!

But all these i was able to find instructions on clublexus. Nothing with manifolds :/

This will give you access to all the Toyota/Lexus factory manuals, wiring diagrams, diagnostic workflows – basically everything a dealership tech would have access to. You'll find all the info on removing the manifolds and installing the new ones, as well as the correct torque specs for all the fasteners. And you can print and save the document as a PDF. I've used these manuals for several repairs before and they've always made the job so much easier – worth the $15 for the correct torque specs alone!
