When playing my radio in accessory mode (engine not running) it turns off after 20 minutes. I owned 2006 GS 300 and it didn't turn off until after a hour of playing. Is there a way to change it from 20 minutes to 1 hour?

I don't think so. The 20 minute turn Off period of course is to help protect the battery from draining down. Apparently Lexus has changed the time period from what it was in '06.
