I would go with the '06 (actually I have the '06 ). These cars are good for more than 100,000 miles, but you can't skip time. Leather, plastic, rubber all ages. Two years maybe not that much difference, but..... Skip the '07, not enough of the return on your investment. Just make sure and purchase extended warranty from lexus - if available (gives you additional 50K and 4 years of almost bumper to bumper warranty - some things are not covered, like electronics)

I do a 5 hr drive each way 8x a year thru desert and high wind mountains (Phx to SD) and the GX is the first car I've owned that I can honestly say I'm not tired when I get there.

As of tonight, I'm a proud owner of a 2006 GS 470
I'll take delivery this coming Friday or next Monday.
Already have a set of Toyo winter tires on the way and tomorrow I'm ordering the Thule box! The third row is indeed a bit tight, but heck, I dont think my 2 year old will mind much in her booster seat!
Life is sweet! Thank you for the input fellas and gals =)

I myself have taken the 4 times on a 7 hour one way trip from LA to SF and SF back to LA and that is the most comfortable ride i've been in. We've rented suburbans and tahoes before but not as comfortable as this.
I do also have the speed regulator (wife) like some of you guys here but I do the driving from 1am and by the time my wife and 15 month old baby wake up in the morning, we're in SF already.
