I would only lock the differential if I feel lack of traction or steering response like you would get on an icy road or going through powder.
It also helps on my driveway - its uphill and I do not plow or shovel.

I just came back from Tahoe, it was snowing for 3 days. You dont need to carry chains if you have 4x4. You dont need to lock the diff unless you stuck in snow. If you stuck in snow just keep your wheels straight.

When driving in light snow. Press the 2nd Start button and leave the center diff alone. If snow gets heavly and you need extra traction to get out a parking space or drive way. Lock the diff! On the hwy with snow you can lock the diff and drive up to 55 mph. Wouln't recommend it. VSC will turn off and I think you would want it on to help driving in snow. Trust the GX electrics nannys help.

There's no need to drive around using 2nd start in light snow or heavy snow for that matter unless you cannot get traction from a dead start and I have yet to experience conditions (driving in 2ft of snow) that the GX cannot handle in "Drive." The GX is all-time 4 wheel drive w/traction control which will limit wheel slip providing enough traction and get up and go in most conditions.
