I'm thinking of getting the TRD 17in matte black wheels for my 2021 GX460. Any recommendations on where to purchase and recommendations on best tire for the TRD wheels.
Also, would I need to make any adjustments or will the new wheels and tires fit on current setup?

I do not believe there is any difference between fitting wheels on a 2019 (mine) and the 2021 but you would want to verify that. If there are no differences then no you will not have to make any adjustments.
Best tire is a completely different animal and depends on YOUR needs. Good thread on tires and wheels and you will find a lot of answers Here
Provided everything is the same between the 19 and 21 these are the part numbers I used for mine, with no issues
TRD Pro black wheels PTR56-89210-F2
TRD Pro Center cap new red ring PT280-89210-F2
Lug nuts black PT076-60200-02
Toyota dealer sale on Black Friday\Cyber Monday 2 weeks after I got my GX and is going to be the best bet, but a ways off for you yet. Total $845.37 all in including tax and shipping for me that was for 5 wheels, 4 sets of center caps and 4 sets of black lug nuts. I'm a big fan of putting a spare into tire rotation. And having a spare that fits in the stock location. I went with Falken Wildpeaks AT3W 265/70r17 and couldn't be happier with them.

I've made a few other changes over the winter as well. Stock with no lift or plans to.

Until I put the sliders on had to pull the infamous front inner fender wheel well bolt with zip ties back. Heat gun didn't hold the plastic. Only happened at full lock as well as going over different heights with articulation. Once the sliders were put on it took care of that altogether.
REALLY tight fit with the spare have to align the lip on the factory hitch with a groove in the tire to get it in. The struggle is real. Any larger even a tenth of an inch it would not fit. The Wildpeaks have gone to the AT4W and have read they are slightly larger and may not fit. One reason I buy 5 tires at a time you can be assured that they will discontinue them on you. Same with decent boots, I always buy two at a time.
TRD wheels are 7 pounds lighter than Lexus OEM so the extra tire weight is offset at each corner. No loss in gas mileage. I've closely monitored my gas mileage after each change- wheels and tires, bumper cut, grille and sliders. It has stayed the same at each step of the way

Although that can change from 19.2 to 19.5 mpg on that 75 trip depending on traffic and wind. Obviously I didn't buy the GX for the mpg my Taco got 16 mpg average for the 11 years I had it prior to the GX. But don't want to circle the drain either LOL.
