I notice this today on my gx. When front window defrog button is pressed (becomes green), it does not kick in ac and fan into high speed if my current mode is manual and fan is set at low speed or when fan/ac is in off state.

Below is a post I saw from a lx470, he described same issue.

"When the front windshield defogger button is pressed the only thing it does is, it directs the airflow to the windshield. However like the other lexus models, it does not turn the fan speed to full, neither does it turn the A/C on.
Is this normal for the lx470 / land cruiser models ?"

I have noticed this. I keep AC off with heat running and when I use defog the AC remains off. Turn off the system and press defog and the AC will run or you can turn it on with the bottom left touch screen toggle.

Nothing to change behavior seen by OP but other HVAC settings one can make with TS.

In auto mode, defog button works as expected.

However in winter time, when I was using manual mode with low fan speed and a/c off, it is a bit confusing, I need to press off button and press front window defog to make fan speed up, and a/c remains off in my case.

So I figure that lexus just wants us to stay in auto mode and void trouble,.
