I am looking at a 2014 luxury with 172k miles. As far as I can tell this the highest mileage 2014+ model for sale in the US. It is a one owner, southern truck, priced at 21,500. I currently have a 2007 gx470 and was looking to upgrade. Does this mileage concern you? Outside of the obvious things to look for (timing cover leak, valley leak, valve cover leaks, ect). I wasn't in the market for a 460 8 months ago, so I'm unsure as well if the price is inflated. Thoughts?
Well I recently purchased a 2014 with 175,000 miles. It was a 1 owner car. I checked the gps history and the previous owner was driving all over the state to golf clubs, so mostly highway miles. He also had his home address programmed which was promptly deleted, but he lived in a very nice luxury high rise in town. So, I thought I scored. The interior was immaculate. The outside had the dents and scratches you would expect with that mileage. A few days after buying, I got a check engine light, which I later discovered the dealer had deleted. I have a long post about it here. But long story short, it is currently in the transmission shop getting a total rebuild to the tune of $3k+.
I believed the hype that these cars are "million mile" tanks and to disregard the mileage. I don't anymore. Caveat emptor applies on these for sure.

Look up the valuation here ... used Louisville Zip Code and 175,000 miles ...
I personally wouldn't buy it at that price. Yes, it's a Lexus but 175K is approaching more than half of its usable life and major repairs can be expected (low probability but it's there). If it's under 20K (18K-19K), I might consider it.
I would pay a few grands more for a sample that is under or around 100K miles. You'd get the Lexus "trouble free" you are looking for.
These rigs are a dime a dozen. Either get one a few years older with less miles for about the same price, or get one of the same year with less miles for a few bucks more.

Do you have a carmax anywhere near? Even a small road trip away? I've purchased cars from them in the past. I also use to run a small used car dealer. It's pretty well known they have some of the highest standards for used cars on their lot. You can have them ship one to your nearest carmax from anywhere in the country. One of my tricks has been shipping cars and saving a couple grand that way. For example I live in Miami. Used Mercedes are in high demand here. I found the exact model with the specs I was looking for several states away in a smaller town. It was selling for 2 or 3k less than the same one would sale in Miami. I only had to pay 2 or 3 hundred to have it shipped here. Totally worth it. Also they offer a really great extended warranty for their used cars which makes buying from them almost problem free. Or as problem free as you can get when you are taking about used cars.
I am not 100% on the build quality diff between 470 2UZ-FE (J120) and the 460 1UR-FE (J150), but my 470 is at 225kmi and the lower end is very clean, little to no carbon on pistons, UOA's are very good, has a bit of tick on the valve train (probably needs shim replacement). I believe my 2UZ-FE can go 300-350kmi, maybe more. I did however just run into a fuel pump issue.
460's also changed transmission unit, so I am not sure how that compares to the 470's sealed "world" transmission.
A 6yr old 460 w/ 172kmi, so it already had two timing belt replacements?, or it will need a 2nd one about now.
If the engine was well maintained it should be ok. It's the peripheral stuff around the engine that may need attention (wheel hub bearings, CV axles, driveshaft, etc etc).

Whats wrong with your current GX470? I simply ask since the used market in general has been high for the past couple months (only trending back to normal maybe a month or so) so you can get out of your current vehicle for more while simultaneously having to pay a premium on the 460.
Price wise its seems high, but whats the pre-purchase inspection at Lexus or a trusted indy say? The seller can say xyz, but that doesnt absolve you out of your due diligence.
Leave the myths at home and take the facts to the bank:
1) High mileage cars are somehow mechanically unworn even if they 100x the mileage. Wear occurs either way when your pushing miles, so maybe there is less powertrain wear but significantly higher chassis wear. It all balances out in the end.
2) GX are built like tanks....so tanks dont need maintenance?
There really isn't a replacement schedule for timing chains and are generally considered life of engine.
That isn't to say something can't happen with chain stretch, defective tensioners and/or guides but that is just a failure that happens or some engines have flaws that require repairs.
