Newbie experience and a question.

I am new to recently acquired 450H.
So on reaching a destination, I put the vehicle in Park and opened the door to step out without shutting the vehicle [start/stop button]. Since the gasoline engine was not running, I assumed that the vehicle is turned off.
Just wondering what would have happened if I did step out with the keys in my pocket and tried to lock the door while the vehicle was still running (read Ready).

Salim, take a look at this thread. By design, no, you're not supposed to be able to lock the RX while the vehicle is running. The key fob isn't necessary to drive off once it's started. So maybe there was intent setting it up that way. If you leave the key fob inside, it's also not supposed to lock. But we have instances where it's happened. More here.
When you walk away from the RXh, running and key fob in hand, it should beep three times when you close the door. Key not detected message shows up on the dash. The beeps are distinct from lock/unlock (if yours is programmed that way). Some use the folding mirrors to confirm the RX is locked (and off). The keyless start and silent engine will take some time to get used to.
