Navigation Screen
Being able to play mp3's (CD-R or DVD) is great and the information the navigation screen displays is also great, but when you drive the mp3 detailed information goes away.
It comes back when you come to a stop. You don't need to put the car in park, but you must be stopped.
Since you are able to view the navigation screen when driving, I'm wondering why Lexus made it so you can't see the detailed song information while driving. If it was strickly a safety issue, then why can you see the navigation map, which requires you to look at the screen.
I also noticed some options are greyed-out when the car is moving and you can't select them.

Bluetooth Phone
The only problem I noticed with using my bluetooth phone is you can't transfer a call to the actual phone when driving, only when stopped.
This means when you recieve (or make a call) everyone in the car can particpate in your phone call, whether you want them to or not.
Again, nothing real major, but I was traveling with a friend from work to visit a customer and he got a cell phone call. Then I received a call and it came through the speakers which made it hard to talk for both of us.

Also, while stopped, you can view the number of the incoming call but once you start moving it disappears! This is a really bad design because now I have to "physically" pickup the phone to view the incoming number before answering the call with the NAV unit

Although I really love my IS350, I'm finding how limited the Nav Screen is while driving.
Example:
Speed Dial Numbers and Names appear when stopped, but the number disappears while driving. When you want to call someone via Speed Dial when driving you only see the name, so if you have entered two numbers for the same person (home and cell) you must rely on the little icons to tell the difference.
Again you can see the full name, but not the number. I don't understand this. Showing you the name, but not the number somehow makes it more safe?

I have my work voice mail number on Speed Dial. When I'm stopped I can enter my security code on the touch screen, but when I'm driving I can't enter my 4 digit security code. I know, this is for safety reasons.
