Which sub/amp combo would sound better?
Infinity Basslink (10" driver, 200w)
JL 10" w6v2 powered by JL 250/1 in a sealed 1.25 cu enclosure)

10w6 all the way, I have never liked any of Infinity sub, even the kappa perfect series. 1.25cf sealed is kinda big for that sub, I think it takes less than that. I know the 12w6 take 1.25cf sealed.

i have a 10w3 in my car... i feel if you are going to spend the bucks for a JL 250/1, you may as well spend another 100-150 to get an amp that could be used in future projects (the 500/1 can easily power 2 10w3's if wired to 2ohms).
if you are putting the JL in a sealed box, the 500/1 will not overpower the sub. mine sounds excellent. not too loud, well balanced.

I have the JL setup you are asking about... It is incredible, and insane... No comparison to the Infinity basslink... Night and day difference... JL all the way...
But you are talking a huge price difference between the two, so it's not really a fair comparison...

you could get a JL 10w0 with the JL 250/1 amp. put it in a sealed enclosure, and it will sound excellent as well. here's a pic of my 10w3 in the corner. i fiberglassed the box myself.
