Now that you cleared it all up, I think you are paying a bit much. I knw it's based on what state your on too so take this with a greain of salt.
I got my IS250 for around $32,500 OTD and mine is an Auto, so that $1,000+ more than yours already, with premium package (HID, ventilated/heated seats), 18" wheel with summer tires and floor mats. I say for a 6-speed manual, you could probably get $1,000-$1,500 off the price.
The Taste of Laxus coupon could only be received by aprticipating in the Taste of Lexus event so if that passed in your area, it's too late to get it. I'm not sure if it's tranferable either.
My best advice to get a lower price is go to a couple of dealerships (within the 50 miles radius) and tell them the price you were offered and have each other beat the price. Purchasing at the end of the month or year is also adviceable as there will be desperate sales people in the dealership trying to move inventory.

By the way, why is your sales tax 7%? Mine was only 6%. The Taste of Lexus coupon is $750 off a new 2007/2008 Lexus. It's same as cash so it is applied to your down payment, therefore helping with your monthly payments.

I agree that it's time to play some dealers off each other, but I would do the whole thing via e-mail, using the "Find A Dealer" and "Contact Dealer" functions on the Lexus website.
This will (hopefully) ensure that you are dealing with the Internet Manager rather than the floor staff, will provide you a written record of any price quote received, and will allow you to rule out in advance any dealers who don't even have a 6MT on the lot (they are pretty rare).
Also, if you haven't yet test-driven the car, I would definitely recommend doing so, to ensure that the 6MT is to your taste.

Sales tax isn't dependant on your state in most cases... its dependant on your location in the state... like in california is may vary from something like 7.5 to 8.75...
