I test drove the 2009 TSX and 2008 IS250. I ended up going w/ the IS. After a month of the new ride, I definitely am happy with the decision I made, no regrets.
Definitely the key difference was the interior quality. Lexus can't beats TSX there.

my previous car was the accord and after i got the IS, yes it feels much smaller, but i've never regretted getting the IS. in terms of visibility when changing lanes, it has always been fine especially with the large rearview mirrors. I like the looks of the 09 TSX, but the 2IS looks much better IMO. =D

Here are my 2 long cents...
I have both cars...there is a difference between both...for once you get more stuff in the 09 TSX vs IS250...TSX like real time weather and real time traffic, you can access the Navi while driving, 30K for a fully loaded TSX, (I know you online its priced for 32K) The leather in the TSX feels more thicker than the wrinkle free flat leather in the IS. Performence wise there about the same, even same MPG. The new TSX looks good (it has to grow on you, specailly in black...wow). The IS does feel tight all around...there is more leg room in the rear in the TSX and the trunk in the TSX is twice as big as the IS. There is a lot less body roll in the TSX. I can keep going. If your not going to consider the IS350 your better off with the TSX, your save yourself thousands of dollars. I know a lot of people say the new TSX looks ugly, but it doesn't...test drive both...but test drive the TSX. Any questions you can PM.

I am willing to trust someone who has owned both the TSX and the IS...I test drove the TSX, it does drive really well and I can't tell the difference between FWD vs. RWD...plus I4 is really quick and smooth, for a moment I thought the TSX had a V6...it is in fact more fun for sure than the IS, BUT the IS seems to be more luxurious, in reality it is the best luxury car in its class and refined second to none. Even the Bimmer begs in front of the lexus in this department.. but for some, some reason, that fun factor didn't kick-in when I test drove the IS, I mean I didn't go saying WOW around..Does anyone feel the same way? I'm going to test drive the AWD version of IS250 now. I think the TSX '08 is a much better car than the TSX '09, including interior, handling and exterior. I'm comparing the IS250 to TSX '08 here.

have old 06 tsx and and 07 is350. the old tsx is actually a lot of fun to play with. we test drove the is250 back then and was only meh in terms of excitement, it was definitely very refined and luxurious inside, but just didn't have it. both transmissions on the tsx are pretty good. the automatic (wife can't drive manual) is actually pretty fun on the tsx. it also doesn't have some of the weird quirks i've come to dislike in my is350; must have key to open rear trunk, can't unlock door from outside with key when power (not ignition) is on, weird inconsistent rattles coming from dash... however, the old tsx nav screen is a bit touchy, alphabet keyboard won't be properly entered when pressed, and resolution sucks. the new one apparently is not a touch screen (i think). of course, it is bigger, cheaper with more standard options, and the fwd doesn't exhibit torque steer like my wagon. it is actually very tossable too. only reason we got the is350 was the 3.5l engine (which is quite addictive hehe) and well, we already have a tsx

I would have to disagree with this. The OP was asking about the 250. You can buy a nicely equipped 250 for about $35K (even less with negotiating a good deal).
Other than the TSX, what competitor is priced less? 3 Series? A4 2.0? MB C300? Volvo S40?

Here are some online prices, I know that the final price would differ so use these as they are.
S40 - $24,365 ($29,325 fully loaded with all available options, lowest trim model though)
IS250 - $30,855 ($34,350 with Luxury Package)
G35 - $32,250 ($34,750, I chose the Premium Package which happens to be the most expensive)
C-Class - $32,975 ($34,725 nicely equipped C300 Luxury Edition)
3 Series - $32,700 ($36,200 with Premium Package)
A4 - $32,700 ($36,700 Premium Plus Package)
Then again when car shopping with AWD and auto in mind, the prices differ and the deal for the IS250 AWD wasn't exactly the best.
