i just got my first Lexus is250 08 im new here at the forum love how you guys helping each other ,want to get your opinion what would be the first mod to do???????

Depends on your budget for your first mod. I always like to do suspension upgrades after a couple of minor cosmetic upgrades. For example, on my car I bought small stuff like LED puddle light bulbs, roof spoiler, and GFX muffler tips. Now I've got H&R springs on order, and an F-Sport sway bar set is next. Following that I'll be looking at new wheels. And after that, I plan to make it sound a bit nicer by adding an intake and maybe axleback mufflers.

is the spring and the sway bar will make a big difference driving it ? stiffer ? what kind of coils?

Well the sway bar will reduce your body roll and if you get coilovers, you can adjust the suspension to make it as soft or stiff as you like

First mod should be a set of springs or coil overs and a set of sway bars. It will be improve your driving experience IMHO. Then you can get your intake, exhaust, JDM visors, body kit, rims, roof spoiler, LEDs etc depending on how much $$$ you want to spend.
