Ok, I have never done this myself on an IS, but I have done it plenty of times on my Saturn and other cars...so let see...
1st of all, ONLY USE API GL-5 fluid when you fill it, that comes straight from a Lexus TSB.
The way to drain and fill the fluid in a manual transmission is very similar to how you drain and fill a differential. There is 1 drain plug at the bottom of the housing, and 2 fill plugs, one on the side and one on top. I tried to include some diagrams of the transmission components so you can see where these plugs are, but I can't get the diagrams to upload properly, sorry....and remember to replace the gaskets as well.
To drain and fill, make sure the vehicle is level, and remove the bottom drain plug to drain all the fluid out. Once the fluid is all drained out, clean the drain plug and re-install it with a new gasket. Then remove both the fill plugs. You should pour the fluid into the top fill plug until it starts to spill out of the side plug. Once it stops spilling out the side, re-install both fill plugs with new gaskets, clean any residual fluid, and you are done.
Again, I have never done this on an IS, so there might be something I am missing, so everyone who has done this please chime in...

quick123 pretty much said it, except break loose the fill plugs before removing the drain plug. it really sucks when you find out your fill plugs are frozen after all the fluid is out. just a tip. and a diy with photos would be great.

I actually had to change the pressure plate and clutch disc on my IS250 since it was worn from the previous owner. In the process, I had to drain and fill the trans fluid. Draining was simple since the drain plug is the only plug at the bottom of the trans bell housing. Should be pretty easy to locate once you're under there.
Sorry I don't have any pictures to post at this time.
The filler plug is on the side of the transmission on the driver side (left side). If I remember correctly, it was 90 degrees from the drain plug in the horizontal position. The following pdf file shows the service level of the fluid. But be warned! The filler plug is in a tight space between trans housing and the body. You will need a pump (manual or otherwise) to be able to pump the fluid into the filler opening.
I got a hand pump from a local tool store and used it to pump the fluid into the transmission. Without this it would have been very difficult to get the fluid into the transmission.

Yeah this will be my next maintenance job I do since I got a bit over 60,000 miles on the IS. Will probably go with Redline Synthetic. Does anyone know how many quarts the bell housing requires?
