Honestly, I rarly go under 3/4 of a tank, as I commute less than 2 miles a day to and from work. The 35mpg I got on the way to Dallas from austin is the best realistic mileage I've managed in my 250MT.
Shui, seems like your mileage on your 250 is fantastic, btw!
Also, fun aside, let's focus on helping the OP with his question, rather than bickering about which method is most accurate... It's agreed that it varies for lots of reasons, but with a loaded car, (family, bags etc.,) a good (conservative) number for calculating fuel cost would probably be around 350 miles per tank. Many here can reach 400, but with a loaded car, some traffic... Just my thoughts.

When I did 10 hours of highway driving once I got 475 miles out of a tank with my IS350. After filling it up I worked it out to about 31MPG. Could have broken 500 easily, but it would have meant dipping into the 2 gallon reserve and I wasn't entirely sure where the next gas station was.

I mostly drive about 65/35 city/highway and normally get around 25 mpg in my 250. Anytime Im on the highway I go 70 and hit the cruise control on.
My most recent tank I went 60 instead of 70 and got 395 miles to the tank (estimated 35 miles left) which was 29 MPG! I noticed that my RPM was right at 2K when I was going 70 and dipped down to around 1.7-1.8 when going 60. I never knew how big of a difference the lower RPMs made. Im estimated to get 450 miles on my current tank but that probably wont happen

Looks like the normal fuel blend has come back. Can finally pull 400 again! Don't worry about the exclamation mark... wants me to change my oil which I will do soon; did not bother to turn it off lol!

Here are my averages on a 2007 IS350 running a catless setup w/ tanabe axleback; my commute is around 70% highway 30% local:
Car's lifetime average: 23.0 mpg
Most recent Tank average: 25 mpg
Most recent range when I filled up: 367 miles
