I attached a video of the noise
https://youtu.be/tpRz-8qMpb8
So there's a click in the rear driver's side. I thought it's the break pads at first I replaced them still there. Doesn't happen if I'm just cruising only under load. Can this be the CV axle going bad? There's no consistent clicking while driving

It's the rear end. It's a 07 is250 85k miles. I'm the second owner all Lexus maintenance history. It just started recently. Doesn't click on turns. Only under load if I accelerate fast or slam the breaks. And only from a stop. Not if I'm moving already. I thought it was the breaks moving around but I guess not I changed them and still the same thing

Ok thanks. So it might be the differential? And if unscrew the pinion but will oil leak out that I have to refill. I don't want to open it up and not be ready and end up having to fill it back up

I said; "check the pinion nut" NOT LOOSEN THE PINION NUT! IF YOU DO THAT, %$@# will hit the fan!!!
Point is; you don't know. So drain the fluid via the provided drain plug. If silver and shinny **maybe** it's the diff. If just dark, probably NOT the diff. They don't just click for no reason.
You do know to fill it with oil once drained, yes? You will need a pump and fluid!
So, if the oil is clean you could have some loose bolts, bad bushings, or perhaps and axle CV joint going bad.

Did you ever figure out what this was? Check out the U-tube video below. It is not a great step by step DIY video but kind of gives you an idea what is involved. One of the rear axles was making the clicking noise.
I have a 2007 IS250 RWD and during my last oil change at 98K, dealer told me both my rear CV boots are torn and splashing grease all over the place. Previous oil change at the same dealer at 95K, boots had no issues. Both boots ( 1 on each rear axle) getting damaged at the same time? I don't track and drive the car mostly freeways 90% of the time and the other 10% is all city driving. Lexus dealer quoted me $4,000 to replace both rear axles (rebuilt ones) with labor. $2,000 was what he quoted me for both axles. Can someone with more experience chime in and let me know if it is OK to drive like this for say about another 5K miles or so if it is only the boots that are leaking grease without major damage? Car drives and handles extremely smooth. No noises etc.
Thanks.
