Thankfully in most cases higher mileage Lexus 2nd Gens that were taken care of usually have solid engines and transmissions. Things to look for:
Create an account on lexus.com and check the VIN for service records, that recall issues were addressed
Check that the VIN is clean and not a salvage, as well as accidents if any
Models 2006-2009 make sure the "Sticky Dash" issue has been addressed. For 2010 models the extended warranty runs out this year
For IS250's, carbon buildup will be an issue on higher mileage vehicles
Water pump and alternator are another failure point at higher mileage vehicles
Have the transmission fluid checked. Although it's a sealed system, it should be swapped out at around 100K
Other common things to check for:
Plugs
Vacuum leaks
Exhaust leaks (rusted pipes, worn seals)
CATS are in working order
Oil leaks and or Oil burning
Others will add more to this list
