Have you tried using Customize yet? I don't have that option in my IS 350, but from what I have seen, you can change various aspects of the driving feel like engine response and suspension independently from one another when using Customize.
As for Eco, it just dulls down the throttle and reduces the maximum output of the HVAC system to save fuel. Sport S and S+ progressively get more sporty in feel, with S+ having a firmer suspension.
