100K miles (or 10 years?) for the 1st change, and then every 50K or 5 years after that.

Engine Coolant
When inspecting, visually check the engine coolant level on the see through reservoir. Add coolant as necessary. When replacing, drain and flush the cooling system and refill with coolant. Always use Genuine Toyota Super Long-Life Coolant or similar high-quality non-silicate, non-amine, non-borate, non-nitrate ethylene-glycol coolant with long-life hybrid organic acid technology (i.e., a combination of low phosphates and organic acids). A qualified technician should perform these operations.
Note: Initial replacement is at 120 months/100,000 miles. Replace every 60 months/50,000 miles thereafter.

Anyone know how much coolant you need to buy to flush the system?

I would flush with distilled water first.

As @Rickna said, flush with distilled water, get a few gallons of those. And you’ll need 2 gallons of pre-mixed pink coolant, and will have a little bit of leftover.

Thanks for the clarification. My question was mainly asking how many gallons you need to buy to replenish what you flush, so roger that on the 2 gallons, and will definitely use distilled water first.

Meh. Flush is a waste of time unless there's corrosion. I've never flushed mine and only added new coolant (like when I did the valley plate). Any distilled will only dilute the premixed coolant you put in there. Not exactly all bad depending on where you live. Running an 80/20 distilled to coolant mix is what I've done in my Supra for a very long time to minimize knock. Water is a far superior liquid for heat transfer, so more of it is just fine unless you live somewhere it gets genuinely cold. 80/20 distilled to Toyota red coolant freezes at 7 or 8F (I tested it) but was able to give me a whole octane point of detonation resistance.
