Wow, that's a sweet offer Lav. I think though he'll need the full dealer version of Techstream to isolate the issue, but it's worth a try with Techstream mini. B1216 is not an OBD II code.

I think, that diagnostic with TIS Techstream will give an answer. This tool could connect with all units in the car. You should check fuses also. Maybe there is no +12V or there is problem with communication with gateway ecu.

Mine is the full version 👍. The only thing it does not do is the firmware updates. You get every other control. ABS, smart key programming, etc.

For whatever reason it reads my vin number one number off. The vin it reads does not even exist. Probably the software. Everything else works. Mine has always worked. I have used it on multiple Toyota and Lexus vehicles. It's good stuff. You can also do data logging of sensors and maps and all kinds of things.

Just swatting at straws but wondering if the passenger side front door control box same as the driver side front door box? If so it might be interesting to swap front door boxes and see if the problem moves. Most likely driver door box is unique though...but doesn't hurt to check.

The ECU we're troubleshooting is the driver-side (RH) rear door, and we know that the passenger-side (LH) rear door ECU is working properly. So, Jabberwock's suggestion is definitely worth a shot...

Attached is the electrical schematic for the wireless locks and windows system. The Rear Door LH (passenger) ECU is shown on page 255, and the Rear Door RH (driver) ECU is on 256. The 2 ECU's are identical except for Pin 15 (Labeled "SEL1") is grounded for the Rear Door RH (driver) ECU (page 256).

also take a look at Post #13 for Inspection Procedures for the ECU.

It would be great if you would report back on whether you solve the door ECU communication to the main gateway ECU using Techstream mini.

A quick update. Mike stopped by today and we checked his car. The left door "ecu" is not communicating with the main computer. The door computer is more like a control module rather then stand alone system. None of the other computers in the car are stand alone.

There is no programming of door modules. The main ECU handles the communication to the various other small computers throughout the vehicle. Really everything happens in the main computer. Smart keys, alarms, user settings, error storage, etc. The other units act like smart switches. Anyway, it is very interesting to look at. Next step would be to check for power to the module and replace module. So yes, it was considered a gateway fault. No communication with the left door ecu.

Thanks for returning to report outcome. Were you able to isolate the problem to the door ECU or the main gateway control computer?

Lav the man! Good work looking for that solution you and Mike.

Maybe, just maybe, an incident where a rat ate off of a wire leading to the ECU?
Happened once with my dad's BMW, took us ages to figure it out. One of the hidden wires that lead to the ECU motherboard was chewed off.

Don't know, but that mouse has bad taste in cars.

It could be the problem with Gateway ECU or with harness between gateway ecu and door ecu.

This schema would be help to understanding comunication routing.

Mike mentioned that the door ecu was replaced by a used model i think from eBay. It never actually worked from what I understand. This is most likely the culprit. It could be the connection, broken wires, missing lead. Heck it might not even be connected. Really have to open the door to check. Could be all kinds of things but most likely it is the ecu itself.

Mike, since you already changed the door ECU with one you have confidence was working before you installed it, suggest you go back to what was originally considered: that the replacement door ECU must be programmed (but can't be by Techstream mini) to communicate with the main gateway ECU. Check with Lexus and/or Toyota dealer.
