Doesn't *Anyone* Want to Sell a Car??!!
Grrr.
I've contacted three different dealers here in NorCal, using personal recommendations for specific salespeople (one at Magnussen's, one at Pleasanton, and one in San Jose).
First person - initial email on May 16th, followup on May 19th, still no answer.
Second person - we've swapped emails, but he hasn't responded since I gave him my "special order configuration" details. So that's been since Friday last week, despite two contacts from me.
The third - hasn't given me her email address, and continues to play voicemail tag. Now she's off today and tomorrow.
Silly me - I thought salespeople wanted to SELL. This is getting ludicrous.

I wouldn't think that salespeople don't wanna sell the car,,,
maybe they are not taking any email request seriously or maybe they want you to come in to the dealer
when I was shopping for my cars, I emailed 10 different dealers (LOL) and only 6 responded back to me

Nope, that was a whine about a clueless local dealer, that my financing company has forbidden me to use anyway.
Yeah, and that's lame. This is a purchase for me, much like any other appliance. It's like going to Amazon and ordering a chrome blender instead of a white one. So why can't I work it out in advance, and then go into the dealership to finalize the details and sign the contract? Oh, that's right - they want to hold me captive for hours while they ask me "What will it take to get you to buy this car TODAY?!"

Yeah, I may end up "defiantly" at the dealer in the end. But they won't like it.
I've contacted them via phone and email, with definite option choices and clearly having done my homework. I know it's going to be a special order; I know what colors, options, and packages I want.
I shouldn't have to waste 3 hours at a dealer when I'm spending $45K in order to be taken seriously.

From having talked with Lexus sales people from multiple dealers, they seem less inclined to want to do special orders then say someone from a BMW dealer where almost anything is possible. I've also been told that Lexus special orders take longer than some of the German makes, though I am not so sure how true that is; this is what a Lexus sales person told me.
With that said, if you're choosing to email people rather than call or show up in person, for something they may see as a lower priority then I'm not so sure what you should be expecting. Maybe if you start your email off as "I'll pay MSRP for..." you'll get their attention, but then again I'm sure you also want the car at invoice price...

I don't want the "car at invoice price."
The conversations on the RX350 subforum say that people have paid anywhere from $2-$4K under MSRP, to $1200 over invoice.
I'm aware that it's a new model, and "invoice plus $1200" is unlikely. I'm not stupid, nor, I think, unrealistic. I'm also clear that I'm likely to pay closer to MSRP for a car that's an unusual configuration than one that's LOADED.
You'd think a polite email with clear specifics, and an offer to come in to discuss it would be sufficient to get some responses...but maybe not.
