stock tranny is known to only hold up to around 350hp. going turbo on a 4xx is going to be really really costly any way you go (TT or big single) since there are no ready made bolt on kits.

and in my opinion, you'd be foolish NOT to build the motor and tranny around it, because a.) youd be limiting yourself in terms of potential power output and b.) the money spent on building motor and trans to match power reliably would not total anywhere near the cost of blown motors/trannies that WILL happen down the road.

if you absolutely cannot afford to build the motor or tranny, be warned, you will only be able to see about/less than 350hp to the wheels reliably.

To "safely" run 450 Rwhp, I would definitely use a standalone computer. You will need to run larger injectors, I am running RC 550cc. Although I also agree that 450 rwhp is gonna be taxing on the drivetrain, I also believe that if you're not always pounding the crap out of the car, the tranny "should" hold. How often is someone gonna be putting that much power out on a regular basis.

If you aren't gonna rebuild the tranny, at least get a tranny cooler even if you don't get a higher stalled TC.

If you look up other GS4xx TT's (flashgordon, cowboy and morris come to mind for me) They will probably tell you that you will be running the edge on stock pistons. The stock piston's weakness is the ring land. Most of the blown piston are due to cracks in the ring land. They are extremely thin. If you have questions on a built setup, I am currently doing one with a running diary. Check it out.

FYI, if you have the fabrication ability and patience, a TT setup is definitely doable without the crazy costs. Just remember that undertaking this venture is definitely not for the weak of heart.

Cowboy was talking about that specific car (AG4's). Cowboy's car is a 1uz tt'd beast tranplanted into a supra. You gotta see that one. He's gone nutz.
