Mr Big,
Probably don't need to, but you can just in case and it won't hurt anything
You can reset it by
a.) removing the battery cable for a few minutes
b.) pulling the ECU fuse in your fuse box located (in the 4) in the engine compartment near the windshield, passenger side
Photo courtsey bananags
And a thread with a decent explanation click here
If you flip open your fuse box and look at the inside of the lid, it will have a map to tell you what each fuse does.
If you have a non-nav car, you will lose your radio settings and your personalized seat / mirror preferences.

Airflow problems on GS 430. Lag, Hesitation
I have installed a k&n air filter in my 2001 gs430. 1 month later i started experiencing problems. When I push the gas under low RPM, the car stalls and RPMS drop, then I'll push the gas again harder, and the car would drive. problem got worse. When my mechanic checked it out, the computer showed that it was the maf. He remove the K&N filter, and it looked like playdough. Very dirty and greasy. I never had that problem before. The ECU light never went on.
I see here that many people complain about the same problem with their GS430S. I dont know what the problem is exactly. Can it be the MAF? or Cleaning the fuel injectors? or the TPS? or throttle body?
Any feedback would help a lot. Thank You.
