Actually, one of the members from Taiwan on either the CL or GS300/400/430 group on facebook posted a few days back about having accomplished this swap.
...and as for the question posted above, I doubt anyone would buy a car (other than a parts car) solely for the transmission.

No, maybe YOU should be to one to re-read. Like I said the first time, I doubt anyone would buy a totally separate car just to have a 6-speed, 8-speed, or whatever-speed transmission that the car comes with, when they could just swap the transmission over. Would you buy a Toyota Aristo or Supra just to have a 2JZGTE? Get my point now?

As for 5at->6at conversion, the new box will bolt on, and engine/trans harness can be modified rather than fully replaced. Immobilizer on both 5a/t and 6a/t LS430/GS430 can be 'fixed' (I've done it quite a number of times when fitting the 3UZ into various makes/models). Lexus may have changed the way engine ECU talks to instrument cluster when upgrading to 6spd, but that would be addressed easily by installing 2004-up cluster with the 6spd ECU. I suppose that the real pain would be making ABS/TRC/VSC run properly, which may end up as either giving up with ABS or transplanting 2004+ ABS/VSC ECU as well as maybe some additonal sensors for it.
I think the only way the transplant is practically achievable is buying a complete wreck and transplanting every bit out of it.
6spd manual might seem to be done easier, but factory ECU isn't happy working without factory a/t. While standalone ECU will not talk to instrument cluster and ABS.
