Through the Lexus Parts at the dealership or better still through one of the CL sponsors,
It is a two volume set and I have heard of additional books which cover electrical wiring etc.
Soft copy is also available, and I don't know if that stuff is genuine or not.

RX330 shop manuals
You didn't specify whether you wanted the RX300 or RX330 manual set.
The RX330 manual set is three main volumes and one electrical volume, and runs around $600.
Note that the main manuals are intended for factory-trained service tech's, so they don't go into great detail (e.g. "Remove rear left underfloor cargo box"). The electrical manual is, of course, very detailed.

I got one on CD from eBay for $20 last month and it covers EVERYTHING. It even details the aerodynamic design and the "hawk-like" appearance of the RX330's headlights. Very interesting to read.
A bit of a pain to print off each page but the pages on the CD can be searched with Acrobat reader by keyword. Appears legit but at $20 it likely isn't ;-).

Toyota has a Web site for accessing service manuals, TSIB's, and other technical info online for Toyota, Lexus, and Scion. It's at http://techinfo.lexus.com/. You need to register and pay for access. The cost ranges from $10 for one calendar day (ending at midnight PST), $50 for one month, or $350 for one year.
The site has everything in Adobe Acrobat (PDF) format. Don't expect to download huge manuals, though. The service manual is cut into hundreds of small pieces; the average piece is about 3 pages, covering a very specific topic. The pages are full of hyperlinks, so when the manual tells you to remove something, that's usually a link to the 3-4 pages about that process.
If you're looking for something specific, the site is pretty handy. (It's as good as the paper Service Manuals, anyway.) If you're just looking for general info, it would take way too long to download and capture everything for future reference.
