99 RX300 - misfire on cylinder 5 - P0305

(I posted this question in an old thread, but I wasn't sure anyone was still reading that thread, so I'm posting it here too. Sorry if this breaks with protocol.)
I'm getting a P0305 diagnostic code - indicating a misfire on cylinder 5. I've replaced all the spark plugs, and thought I'd moved the ignition coil to a different cylinder (in the front - where it's easier to get at it). But I'm still getting the same problem after all that work.
So, what cylinder is #5? I had assumed (I know, this was my biggest mistake) that they were 1, 2, 3 (left front to right front), and 4, 5, 6 (left rear to right rear) - which would have put cylinder #5 in the middle-back position (the hardest one to get out).
Is it possible that they are numbered 1, 2 (left side, rear and front), 3, 4 (middle rear and front), 5, 6 (right side, rear and front)?
Also, what else could cause a misfire on this specific cylinder?
Lastly, what harm is this doing to my engine to run it with this misfire (I haven't been driving it a lot, but I did take it to the auto parts store to hook up their diag-tool after I replaced the spark plugs).

Our's is a sidewinder.
From the front of the engine .... looking at the engine from the right front fender looking across the engine towards the air cleaner... left front is # 1 Right front #2 Left middle #3 Right middle #4 Left rear #5 Right rear #6
Standing at the radiator: Closest to you are cylinders 2 4 6. Closest to the firewall.. 1 3 5 Left to right

Maybe something in the wiring. I did the same thing you did when I had my problem. I don;t have my service manual in front of me, but I also remember something about checking some connections by the ECM on this.

What a PAIN in the arss to change out an
ignition coil from the back...holy crap and
it's "supposed" to be not that hard given
it's #5.
Ohhhh loosening up the nut was pc of cake
and taking out the coil..no problemo but
try taking out the CLIP on the damn thing
GOOD LUCK
I tried every conceivable way I thought of.
Holding down the clip in the middle to let
it slide off and doing so with one hand, because
that's all you can fit in that space, Forget it!
Nut on or off....left hand or right...WD40 to have
a little slide action...no good..wiped it off to slippery.
If anyone has a better suggestion...I'm all ears
Just thought of an idea...maybe it'll work.
Small...very small C-clamp to hold the clip down and
THEN try to work the wire off....sounds ingenious enough
