When it comes to gear oil [for lubrication or coupling], the bye product are metal particles/shavings. For engine oil you get combustion bye products in the oil. So for engine oil, you should get the maximum oil out by volume. In case of transmission-gear_oil, you should get the sediments out. I would suggest that drain it out by removing the drain-plug.

I'm hoping I'm OK with just using a pump to get the fluid out thru the fill hole. Too much trouble this time around to get that drain plug out. I stuck a portion of a clothes hanger into a tube so that I could bend it into an "L" so that I can make sure that the end of the tube goes into the bottom of the diff. I didn't get to clean off the drain plug but will have to try some other time.

I'm not sure what's up with you Enz but I'm trying to help the board. I spent weeks researching and determining the best way to change the diff fluid on RX300. Talked to five separate mechanics. Talked to a few who used the wrong wrench and/or technique and screwed up their diff. It is absolutely necessary to modify a wrench to remove the front diff drain plug. You don't have to use the wrench i recommended but you will have to grind down a box wrench to do it correctly. It took 20 mines to grind down the wrench. I'm not sure why this is an issue. I now have a tool for life and was also able to help two other 1st gen owners do their diffs also. If wedging in a block of wood worked for you, that's truly wonderful for you. I absolutely wouldn't recommend or risk this when there is a clear step by step solution between what Lexmex and I have detailed.
