I am curious about the need to replace spark plug wires on my 01 RX 300? I have 43k on my baby. Are there any with better quality? Should I consider replaceing them, and where can I find a set?

You should have a distributorless ignition with coilpacks mounted above each sparkplug..
Change plugs at dealer recommended intervals and the coil packs should be good for at least 200,000km

Some of the 1MZFE engines used in certain Toyota models older than our RX300s have an ignition coil (3 coils) and plug wire (3 plug wire systems).
When I was looking a few years back (some may remember the posts) I was looking to see whether the RX300 followed this system or had all coils. Reason was my cousin really enjoyed the Nology wires that I bought him, that burned through the bad Premium gas that existed at the time. I also noticed the difference too when driving his Golf VR6.
I had one coil go out a year ago (Ignition coil 1, front of engine passenger side) after doing some throttle body cleaning. In a pinch (since no Lexus here) I had to buy one for a last generation Camry (not the current one), sand down the sides and it fit like a charm (don't do this except in an emergency) until I could get the part shown below, which I keep as an extra handy.
90080-19016 is the latest part number for the RX300. That part number differs from the OEM, but it will fit. From what I remember, each coil costs a tad over $100 USD in the U.S.

Man, I love this site, I learn something new every time I log on.
No sparkplug wire at all, that's pretty advanced! My C5 Corvette has a coil
per cylinder but it DOES have a short wire.
Lexmex has really been through everything on his RX!

Just a quick note. Several Toyota models (highlander, avalon, etc) use the same ignition coil (part # 90080-19016) as the RX300. I found one online at a parts store for $25 Canadian. Saved me from my mechanic who asked for $235 Canadian for the exact same part. I've seen several go on e-bay motors for 12$ or less. Just keep your eyes open on the web b4 going ahead with the dealership.
