NO, there is no audio no matter what. If I play CD I can see time is running I can change songs, but there is no audio. Seems like volume control is not working.....

I thought I'd wait for somebody else to comment, since I have no first-hand experience with Lexus audio systems. However, I think that the two major modules are the head unit in the middle of the front dash and the amplifier. (I also don't have my maintenance manual with me; I'm in Florida and the manual is up North).
I think you need to start troubleshooting by seeing if you have audio on the wiring between the head unit and the amplifier. Also, I'd check to see you have 12v power at the amplfier. I'd suspect power/power supply issues since both channels of the audio aren't working.
In looking for something else, I see that several auto recyclers are parting out RX300s on eBay and selling entire center dash assemblies for about $100-$200. Presumably amplifier assemblies are even less expensive. At those prices, it doesn't pay to try to repair; just replace.

I have the entire electrical section out of an 03RX300 might be similar. But I cant attach it since its larger than allowed to be posted. I can try to post some by typing some of it out. You live here in NJ where by are you? I also have my spare center console we can just take yours out and try mine in for a sec to see if its the center console, if that doesn't work them possibly your amp is gone. Its up to you, I can try to help this way. Let me know Julius1980.

You too?!
Hey Bob!
My friend has the same problem w/ her 2000 rx300, which we just started to diagnose today.
I was wondering if your audio select dial is functioning, on her car, it will select bass, treble, etc... but will not change the value.
Anyway, I' think when it stops raining, I'll look into this more, but was wondering if you found anything more...
Thanks

Don't know if this helps but attached is an audio system wiring diagram for a 2000 RX300.
An issue, apparently rare, that can cause the audio system to mute is if the "tele-mute" wire (labeled TMUT on the diagram) becomes permanently grounded. Based on what I have read on this and other Lexus forums, this problem is more likely to arise if the car has or had a dealer installed Lexus phone system. The problem can be caused by a defect in the phone system even if it is no longer used or if the phone system has been removed by cutting wires rather than unplugging the connectors on the factory installed phone harness to the dealer installed phone system components. I saw one thread about the TMUT wire getting grounded from corosion caused by water getting to the connections on the amp.
That said, I think it more likely a problem with your amp or headunit.
It might be worthwhile to have a technician evaluate the problem. The tech at my local indie Lexus repair shop seems to be able to debug audio system problems - at least to the extent of determining which component is causing the problem so they can send it out for repairs.

Nice
That Diagram is BEAUTIFUL!
I have to replace my speakers in my newly purchased 01 rx300, It only has 56000 miles but someone liked their music as loud as I do. It is the pioneer setup. It looks to me by this diagram the amp powers all the speakers including the sub, Is that the case? Is the sub under the passenger seat?
Thanks guys...

I found out that my head unit in the middle of the front dash is working fine, so now I think I have to find out how amplifier works.
Somebody could tell me where amplifier is located?
