2002 RX300 2WD - 125K miles, no major issues thus far BUT I have the following indicator lights that just came up:
CHECK ENGINE / VSC / TRAC OFF
the user's manual all indicate the same, contact your Lexus dealer. Should I brace myself for the worst?

Autozone and as Lex said, most auto stores will pull the code for free. The VSC and TRAC OFF are often accompanied with the check engine light. The reason I've been told is: The computer will auto shut off the VSC and TRAC off to protect the vehicle/driver/passengers. Whatever is tripping the CEL may adversely effect the VSC and TRAC.

A quick check before bringing the car to autozone to pull the codes
would be to check the vacuum hose is still connected to the air filter.
This happens quite often to my RX, I simply push the rubber hose
back into the metal tube sticking out from the air filter, then
disconnect/reconnect my battery. Everything clears up after that.

That is a very GENERIC combination of lights and MAY HAVE NOTHING to do with any of those things. Many different things can trigger that combination of lights. More of a "hey, you MIGHT want to take a look around" than it is a red flag. Don't run any RED lights getting to the dealer! Do a search on that combination of lights and you'll have plenty of reading to do. 1st check what Lexrally said, but don't lose any sleep over it.

Every time this has happened to me (i.e., Check engine light indication), it has been for one of the air/fuel or oxygen sensors burning out. Each of the three sensors has been replaced once and one has been replaced twice. So, if I were to guess, that would be my best guess. Not really cheap, but not really expensive. Maybe $100 if you do it yourself and up to $500 if you go to a dealer.
