I've driven both. Bought an MDX. Both my wife and I did not like handling and lack of power in the RX. Now this is not slalom handling but just overall feel of the car. Center console made the car feel like a minivan. I realize there are pros and cons to this config but we prefered the MDX. The MDX is wider and takes a little more getting used to but it is not that big of a deal. We use the third row seats periodically. When we have to transport kids or smaller adults, it has been great. So much easier than having to take 2 cars. The RX does have a bit more solid and luxurious feel but you'll have to weigh what is more important to you. This difference was not enough to bias us towards the RX.
As you can tell, there are people on both camps. I don't drive it everyday since it is my wife's car. She could not be happier. I prefer my GS but I don't really have any major issues when I do drive the MDX. When we bought it, the Nav was supposed to be better on the MDX. I've never used the Lexus Nav so I cannot confim or deny this. I just know that the Nav has been great.
Drive both and just go with your gut on what you want and need. None of the differences I mentioned were major. It is not like the MDX is greatly superior or vice versa. I can see it go either way. We just made an assessment of our needs and the MDX edged out the RX. Your needs may mor may not swing the other way. It is all personal. (I have not driven the RX330, so this is only a comparison of the RX300 and the MDX.)

Last year I did the same thing. Drove both and bought the MDX. But after I drove the 330, it was no comparison.
If you like the MDX save yourself some money and get the Pilot, same level of "luxury".
