I wasn't sure on this model if charcoal canister was in the back like on RX330 or not but since it's in the back maybe a fogging machine may be useful to find leaks.
Drawing and info I posted above is specific to the RX300 and the dotted line covers the components in question for this code P0440.

Margate330, thank you very much for sharing. After clearing the air filter box I disconnect and then reconnect the battery terminal and drove 3 short trips (all in a mix of local streets and 55-mile limit) highways in the past week, with each about 10 miles. The first 2 finished without the CE light coming up. A couple of miles into on the 3rd trip, the CE light came back again. (I guess the air filter cleaning was not a right "corrective" effort for P0440.) I tried to identify the charcoal canister but it is not obvious to me where it is located. however, I checked the (most) hoses in the engine compartment a couple of times and did not see signs of leaks and cracks.
In additional to locate the Charcoal Canister and check on its tube connections, I will try to check the connections at the "back end" related to the fuel tank.
I do have a question with the fuel tank filler cap: as I turn the cap loose there is no hissing sound to indicate vacuum or pressure - just silence. I guess in normal working conditions there should be a hissing sound as one opens the cap - I did not pay much attention to in the past when filling the gas tank!
The filler cap is still the one since I purchased the RX300 in 2006. Perhaps the time has come to replace it?
It has been slow on my part but I will keep on it and report back.

Replaced the gas fill cap with a new one and the P0440 code came back after a reset (disconnecting the battery) and drove about 15 miles. Removed the plastic cover for the canister and I don't see any cracks on the hoses:
From the posts I guess "smock test" must be done at a shop? Is my this problem heading to something on top of the gas tank?
