Back Hatch "Door Open" Chime
Today I have to bring some 10 foot boards home from the Lumber Store.
Having tied down the hatch (bungee cord) on top of the boards which stuck out the back end of the hatch, I was not happy to find out that the "door open" signal stays on. It does not stop after a minute or two, like the "seat belt not in use" warning. (It only stops if you shift into Park.)
Is there any way around this? Fortunately my ride was short, but it's a little annoying. Does anybody know if the switch that the door hits can be bypassed or can be pressed into the off position while the door is still open?
. . . and I thought I had no complaints about this wonderful car.

Because this could represent a safety issue (CO in the cabin from exhaust), I rather imagine that chime cannot be disabled. Lexus tends to be a bit of a nanny on these kinds of issues.

Oh. I forgot about the CO problem, but this is the first car I have owned with a one-piece hatch that is hinged at the top.
Maybe that annoying noise is not so bad, considering the alternatives.

Hauling for short distances with the hatch open or partially open is OK, IF you keep the other windows open to prevent CO as much as possible from being drawn back into the cabin.

I would be VERY CAREFUL about leaving any windows open and driving with the hatch open. Because of the aerodynamics, most cars will create a suction out of the windows while you are driving with the back open. I am not sure if the RX does that or not, but I know for a fact that your standard suv will!
If you have to do that again, the best thing is to close all the windows, and put the ventilation fan on high, making sure it is set to receive outside air, and aim the vents at your face!
If it were me, I would use the luggage rack and some ropes at both ends to the bumper area. You could probably carry 20' long stuff if you tie it down right.

Out of Warranty
Considering the litigious nature of our society, ready to file a lawsuit at the first opportunity, no manufacturer is going to provide a way to circumvent a warning or safety-related system that is either cheap or easy.
Maybe try earplugs?

This should turn off the seat belt chime. Of course, it is highly recommended to actually use them when driving! But the way Toyota implemented it, it is truly obnoxious!
Solution:
1. turn the key to the ON position, foot off the brake
2. cycle the odometer with the trip reset button until the ODO is displayed (not trip A or B)
3. turn the ignition to the LOCK (OFF) position
4. unbuckle the driver's seatbelt if it was buckled.
5. turn the key to the ON position. when the dash lights have come on, press and hold the trip reset button and count a slow 12 seconds. (editor's note, hold it more like 30 sec)
6. while still holding the trip reset button, buckle the driver's side seatbelt.
7. the odometer display should read "b ON" or "b OFF". toggle the beep OFF with the trip reset.
8. the process should be completed when you unbuckle or turn the key off.
