Two major fluids that you need to change on regular basis are oil (5000k for convention, 10-12k for synthetic) and transmission (30k or so). Both are very simple things to do, especially on Lexus/Toyota. Other fluids (cooolant, differentials, etc..) are a bit harder to change, but you don't need to change them very often.

The way I see it, DYI for simple things and take a car to a mechanic for something that exceeds cost/benefit ratio.

Is there any instructions on how to change transmission oil?
How about brake fluids?

I have a set of instructions for Toyota 4Runner (my other car) and while several pictures might be slightly different, the procedure is exactly the same. Both transmission fluid changes and brake flushes should be pretty much identical on 4Runner and RX330 (both transmission drain plug is location and brake fluid lines are in pretty much the same place and you can't miss either one of them).
