You might try re-setting the TPMS. The methodology is described in your manual (mine is in the car downstairs, so I am writing from memory). I believe the button to do so is at bottom of glove-box.

My TPMS warning light has been coming on randomly; putting the ignition in the ON position than holding the TPMS reset button (under the steering wheel) for 10 seconds resets the TPMS system.

When I got my RX, the TPMS light came on first evening. I checked psi and realized the psi's were set +/- 26-27. Obviously drop in night temp. caused the drop in pressur ethat caused the TPMS light. That is when I had read about the 're-set'. I would probably try first to re-set TPMS with the tires at a lower pressure - e.g. 26 - then re-inflate to correct pressure and see if that resolves the issue. Otherwise, and I am sure you know this as well as I do, the TPMS uses a battery in the rim/sending part. I have no idea how one checks on the state of that battery. How old is your car? [They say the batteries last 5-7 years, and TPMS has not been around that long?].
Of course, random events are tough to analyze. If not sending unit, then receiving unit? Is thee a specific fuse to take out and replace?

You can thank Ford and Firestone for making TPMS mandatory on all cars 2009 and newer. If one of the TPS is going bad (5-7 yrs old) it is a good idea to replace all 4 TPS. The reason being is each one has a unique IP address that needs to be synced to the on board computer. The dealer typically charges $100 for this service regardless if it is one TPS or 4. Therefore, replace all 4 TPS at the same time. The downfall is it will cost for $300-500 for parts + service.

+1 to that (no TPMS on the 2006). It would only make my winter tires on dedicated Lexus rims that much more expensive (having to buy TPMS sensors for the extra rims). The TPMS system, while helpful, should not replace regular checks of your tires with a proper tire gauge.

you might want to check your spare tire. I had the same problem 2 weeks ago. after taking it to the dealer-they said it was the spare tire that was causing this -since it too has a TPMS

On my RX330/06 about every 30 miles or so my low Tire Pressure Warning Light comes on followed quickly by the Parking brake light, Trac Control, VCS, ABS, and Skid. Have checked all 5 tires and reset TPWL and it's good for another 30 or so miles.

Warning Lights
If your getting all of the warning lights. You have more than a TPMS problem. The only time I get this to happen is if the gas cap is not on tight.
If that doesnt fix it. You will need the error codes read.

mine came on too, checked the PSI, and all were at least 30lbs... then the other day, wife noticed that one tire was completely flat (had a nail in it).
I would have replaced the tire myself but I was so ill that day - called Lexus to fix it. they sent someone out to swap out that tire for the spare (free of charge too!)
however, I now need to take it in to have it rebalanced and get a new spare ~$250! ($180 for a new tire, $70 for the balancing)

I just bought some new tires for 07rx350 - firestone destine s/t. Left car overnight to see if tires were leaking (most likely valve stems he says). Dealer says valve stems are rusted (~33k miles)...not sure if they can get TPMS out. I did not have any lights on dash before bringing it in. Anyone hear the same? results? ideas? just preparing myself for tomorrow. Thanks.

I had one tire leak at the valve stem and the dealer replaced the TPMS sensor for one tire as well at no additional charge. Turned out well.
